A shift in the hepatic immunological pattern from predominant adaptive immunity to innate immunity compromises tissue-specific immunity and leads to HCC

چکیده

Abstract The incidence of hepatocellular carcinoma (HCC) associated with non-alcoholic fatty liver disease (NAFLD) is growing because the global consumption high fat, sugar or Western diet (WD). Current data suggests a correlation between hepatic inflammation and progression NAFLD to HCC. However, contradictory reports on identification specific inflammatory cells cytokines hinder further progress in field. Such controversial are due highly reductionist approaches getting lost details. By using flow cytometry, we have demonstrated that immunological patterns were inhibition HCC, which can be discovered through detection proportion immune cell types functioning as network. cytometry utilizes limited number markers hindering comprehensive patterns. Here, perform scRNAseq collected from mouse model (DIAMOND) being WD prior after development well control mice chow (CD). We demonstrate alters pattern by dominating innate immunity, M1 macrophages, over adaptive B T cells, activation fibroblasts. Each created distinct collective function beyond its cellular constituents. Also, shifts compromised system, transforming anti-tumor carcinogenesis-promoting process. Our suggest discovery modulation could offer understanding curative treatment for This work was supported Office Assistant Secretary Defense Health Affairs Breast Cancer Research Program under Award No. W81XWH2210793. Adjuvants of Immunity: Harnessing Innate Immunity to Promote Adaptive Immunity

Since Jules Freund reported that crude mycobacterial extracts greatly promoted immune responses to antigens (1), the use of adjuvants has become a widespread, but poorly understood practice to promote T and B cell responses (2). Recent studies have begun to identify the chemical nature of several adjuvants and the cellular and molecular mechanisms of their long-elusive immunologi-cal effects. F...

Immunological memory in innate immunity.

‘trained immunity’. Trained immunity provides protection against reinfection in a T-/B-cell-independent manner, with nonspecific potentiation of inflammatory reactions and antimicrobial mechanisms playing a central role in these effects [4] . The current issue of the Journal of Innate Immunity presents a series of studies that shed additional light on important aspects of the adaptive propertie...

Innate and Adaptive Immunity to Mucorales

Mucormycosis is an invasive fungal infection characterised by rapid filamentous growth, which leads to angioinvasion, thrombosis, and tissue necrosis. The high mortality rates (50-100%) associated with mucormycosis are reflective of not only the aggressive nature of the infection and the poor therapeutics currently employed, but also the failure of the human immune system to successfully clear ...

Dendritic cells: translating innate to adaptive immunity.

The innate immune system provides many ways to quickly resist infection. The two best-studied defenses in dendritic cells (DCs) are the production of protective cytokines-like interleukin (IL)-12 and type I interferons-and the activation and expansion of innate lymphocytes. IL-12 and type I interferons influence distinct steps in the adaptive immune response of lymphocytes, including the polari...

TWEAK Attenuates the Transition from Innate to Adaptive Immunity

Innate immunity is the first line of defense against infection, protecting the host during the development of adaptive immunity and critically affecting the nature of the adaptive response. We show that, in contrast to tumor necrosis factor alpha (TNF-alpha), the related protein TWEAK attenuates the transition from innate to adaptive mechanisms. TWEAK-/- mice had overabundant natural killer (NK...
